目的优选离子交换树脂纯化黄连生物碱的工艺条件。方法以表小檗碱、黄连碱、巴马汀、小檗碱含量为指标,考察D113型、D001型、D151型、110型、732型阳离子交换树脂纯化黄连生物碱的效果及优选最佳树脂纯化工艺条件。结果 D151型离子交换树脂比其他树脂具有较优的吸附及洗脱参数;最佳纯化条件为:样品溶液浓度(以生药计)为0.1 g·mL-1、pH=9,流速为3 Bv/h,洗脱剂为40%乙醇酸性溶液(含10%醋酸),用量7 Bv,洗脱温度35℃。结论黄连提取液过D151型离子交换树脂柱前后各成分转移率均在95%以上,纯化效果好,适用于黄连生物碱的精制纯化。
Objective To optimize the process conditions of purifying Coptidis alkaloids by ion exchange resin. Methods The contents of berberine, berberine, palmatine and berberine were taken as indexes to study the effect of optimizing the alkaloids of Rhizoma Coptidis D113, D001, D151, 110 and 732 and optimize the optimum resin Purification process conditions. Results The results showed that the optimum conditions for the adsorption and elution of D151 ion exchange resin were as follows: the concentration of the sample was 0.1 g · mL-1, the pH was 9, the flow rate was 3 Bv / h, eluent 40% glycolic acid solution (containing 10% acetic acid), the amount of 7 Bv, the elution temperature of 35 ℃. Conclusion The extraction rate of each component of Coptis chinensis extract before and after D151 ion exchange resin column is more than 95%, and the purifying effect is good. It is suitable for the purification and purification of Coptis alkaloid.
