从茎瘤芥胞质雄性不育(cytoplasmic male sterile, CMS)系线粒体cDNA中扩增获得了 CMS相关 T基因的2个不同转录本, 分别命名为T1170和T1243. 序列分析表明, T1243是一个保留了内含子的转录本, 该内含子具备Ⅱ型内含子基本特征. 这 2 个转录本是 T 基因选择性剪接的产物. RT-PCR 分析表明,在苗期, T 基因转录水平的表达以 T1170 转录本为主; 随着发育时期变化, 该转录本表达逐渐减少, 而另一个转录本 T1243 表达丰度增加; 到盛花期, 该基因的表达以后者为主. Northern 杂交验证了这一结果. 推断茎瘤芥胞质雄性不育性和 T 基因转录后选择性剪接有关.
Two different transcripts of CMS-related T gene were amplified from mitochondrial cDNA of cytoplasmic male sterile (CMS) lines and named as T1170 and T1243, respectively. Sequence analysis showed that T1243 was a conserved Intron transcripts, which have the basic characteristics of type II introns, are the products of alternative splicing of T gene.RT-PCR analysis showed that at the seedling stage, the T gene transcription level The expression of T1170 was predominant in the transcripts, while the expression of T1243 in the other transcripts was decreased with the developmental stages, while the expression of T1243 in the other transcripts was increased. As a result, it is concluded that the cytoplasmic male sterility of the tumorous mustard is related to the post-transcriptional alternative splicing of the T gene.
