选取40只雌性SD大鼠,分为4组,每组10只。除正常组(A组)外,模型组(B组)、干细胞移植组(C组)和仙灵骨葆组(D组)均灌胃维甲酸70 mg/(kg·d),连续灌胃14 d,A组用等量无菌水灌胃。B组注射生理盐水,C组尾静脉注射2×106脐带间充质干细胞,D组灌胃20 mg/kg仙灵骨葆。给药21 d后,测量各组大鼠血清钙离子和磷含量的变化以及ALP和StrACP水平,剥离大鼠右侧股骨并称量,双能X线骨密度仪测量右侧股骨骨密度,左侧骨切片进行电镜观察结构。结果表明,维甲酸诱导的大鼠血清钙离子、磷含量及ALP活性比正常组显著升高,C组和D组治疗后比模型B组有所下降;B组StrACP活性较A组高,C组与A组无显著性差异。B组骨湿重比正常A组显著减少,C组和D组骨湿重与A组无显著性差异;造模后B组骨密度显著下降,给药后C组和D组骨密度与A组无显著性差异;B组骨胶原纤维排列杂乱无序,结构松散并呈丝状,C组与B组相比,骨胶原纤维排列呈束状,走向一致,排列规则、紧密,恢复到与A组骨胶原相近形态和排列。结果提示,注射脐带间充质干细胞对改善维甲酸诱导的大鼠骨质疏松有一定的治疗效果。
Forty female SD rats were selected and divided into 4 groups of 10 rats each. Compared with normal group A, the wet weight of bone in group B decreased significantly, but there was no significant difference between group C and group D. The bone mineral density in group B decreased significantly after modeling There was no significant difference between group B and group B. Collagen fibers in group B were disordered, loosely arranged and filamentous. Compared with group B, the collagen fibers in group C showed a bundle with the same direction and arranged regularly and closely, A group of collagen similar morphology and arrangement. The results suggest that injection of umbilical cord mesenchymal stem cells to improve retinoic acid-induced osteoporosis in rats have a certain therapeutic effect.
