目的:观察榄香烯联合高温对人肺腺癌A549细胞的协同杀伤作用。方法:用MTT法检测高温处理或常温处理细胞对榄香烯的敏感性。采用15μg/mL榄香烯或60μg/mL榄香烯联合42℃高温处理1.5h或常温处理,使用流式细胞仪检测细胞周期,透射电镜观察细胞形态学改变。结果:高温作用后,榄香烯对A549细胞的IC50从35.43μg/mL降低至28.68μg/mL;MTT法检测显示低浓度榄香烯联合高温对A549细胞具有明显的协同杀伤作用,细胞呈现凋亡形态,可见凋亡小体,同时有细胞坏死。结论:低浓度榄香烯联合高温对A549细胞有协同细胞毒效应。
Objective: To observe the synergistic killing effect of elemene combined with high temperature on human lung adenocarcinoma A549 cells. Methods: MTT assay was used to detect the sensitivity of elemene to high temperature treatment or normal temperature treatment. The cells were treated with 15μg / mL elemene or 60μg / mL elemene combined with high temperature of 42 ℃ for 1.5h or room temperature. The cell cycle was detected by flow cytometry and the cell morphological changes were observed by transmission electron microscopy. Results: The IC50 of elemene on A549 cells was reduced from 35.43μg / mL to 28.68μg / mL after high temperature treatment. MTT assay showed that the combination of elemene at low concentration and A549 cells had synergistic cytotoxicity and the cells showed apoptosis Death morphology, visible apoptotic body, at the same time cell necrosis. Conclusion: Low concentration of elemene combined with high temperature has synergistic cytotoxic effect on A549 cells.
