对油菜花粉总脂肪酸部位的提取(CO2超临界萃取)和纯化(醇洗除杂)工艺进行优化。通过星点设计-响应面法,以脂肪油的得率为指标,对油菜花粉总脂肪酸部位的CO2超临界萃取工艺进行了优化;通过正交设计,以总脂肪酸(亚麻酸酰胺、亚麻酸甘油酯、亚麻酸和棕榈酸)含量为指标,对提取物进行醇洗除杂。优化得到超临界萃取工艺为萃取温度60℃,萃取压力35 MPa,萃取时间3 h,纯化工艺为醇浓度80%,醇倍量为50倍,醇提时间1.5 h。油菜花粉总脂肪酸部位经提取纯化,质量分数能达到60%左右,且该工艺稳定可行,适于工业生产。
The extraction of total fatty acids from rape pollen (CO2 supercritical fluid extraction) and purification (alcohol washing and impurity removal) were optimized. The supercritical carbon dioxide extraction process of total fatty acids in rape pollen was optimized by the method of apical design-response surface methodology with the yield of fatty oil as the indicator. The total fatty acids (linolenic acid amide, linolenic acid glycerol Esters, linolenic acid and palmitic acid) content as an indicator, the extract was washed with alcohol miscellaneous. The optimal conditions for the supercritical extraction process were extraction temperature 60 ℃, extraction pressure 35 MPa and extraction time 3 h. The purification process was 80% alcohol concentration, alcohol times 50 times and ethanol extraction time 1.5 h. The rape pollen total fatty acid fraction was extracted and purified, the mass fraction can reach about 60%, and the process is stable and feasible, suitable for industrial production.
