目的:探讨食管鳞状细胞癌(食管鳞癌)组织中肿瘤相关蛋白表达与临床病理变化的关系。方法:采用免疫组化法检测679例食管鳞癌组织中细胞骨架结合蛋白Fascin、突触蛋白Syn和肿瘤抑制基因P63等23种肿瘤相关蛋白在癌组织中的表达,分析上述蛋白的表达与临床病理指标(淋巴结转移、分化程度及临床分期等)的关系。结果:23种蛋白中,食管癌组织中阳性表达率较高(≥80%)的蛋白是AE1/AE3、P63、CK5/6、CK、Ki67和MUC1,较低(≤30%)的是CD56、CTTN、Syn、S100、CgA和CK20。其中,Syn在淋巴结有转移组织比无转移组织表达升高(χ2=4.167,P=0.041),免疫蛋白CD56在低分化组织高于中高分化组织(P=0.017),细胞增殖抗原Ki67在中晚期组织高于早期(χ2=6.596,P=0.010);而与此相反,P63则在淋巴结无转移时表达上升(χ2=5.095,P=0.024),细胞黏附因子E-cad、Fascin、Ezrin、PLK1在中高分化组织中表达升高(P均<0.05),酪氨酸磷酸化蛋白CTTN和Vim在早期组织高于中晚期组织(P均<0.05)。结论:Syn、CD56和Ki67高表达可能与食管癌预后差有关;而P63、E-cad、Fascin、Ezrin、PLK1和Vim、CTTN高表达可能与食管癌预后好有关。
Objective: To investigate the relationship between tumor-related protein expression and clinicopathological changes in esophageal squamous cell carcinoma (ESCC). Methods: Immunohistochemistry was used to detect the expression of cytoskeleton binding protein Fascin, synapsin Syn and tumor suppressor gene P63 in 679 cases of esophageal squamous cell carcinoma. The expressions of these proteins were compared with those of clinical Pathological indicators (lymph node metastasis, the degree of differentiation and clinical stage, etc.) relationship. Results: Of the 23 proteins, AE1 / AE3, P63, CK5 / 6, CK, Ki67 and MUC1 were higher in esophageal cancer tissues with the highest positive rates (≥80% , CTTN, Syn, S100, CgA and CK20. The expression of Syn in lymph node metastasis was higher than that in non-metastasis tissue (χ2 = 4.167, P = 0.041), while the expression of CD56 was higher in poorly differentiated tissues than in moderately well differentiated tissues (P = 0.017) In contrast, P63 was up-regulated in the absence of lymph node metastasis (χ2 = 5.095, P = 0.024), and the expressions of E-cad, Fascin, Ezrin, PLK1 The expression of tyrosine phosphorylated protein CTTN and Vim were higher in the early stage than that in the advanced stage (all P <0.05). Conclusion: The high expressions of Syn, CD56 and Ki67 may be related to the poor prognosis of esophageal cancer. However, the high expressions of P63, E-cad, Fascin, Ezrin, PLK1, Vim and CTTN may be related to the prognosis of esophageal cancer.
