目的建立了固相萃取HPLC测定大鼠血清中黄芪甲苷含量的方法,比较黄芪总苷(AST)、黄芪甲苷(AST-Ⅳ)在大鼠腹腔注射给药后相同时间大鼠血清中AST-Ⅳ的含量。方法分别以AST、AST-Ⅳ(按AST-Ⅳ计20 mg/kg相等剂量)二种药物大鼠腹腔注射,给药后30,60,75,90,105,120 min测定大鼠血清中AST-Ⅳ的含量,并对两种药物结果进行分析、比较。结果在大鼠腹腔注射等量AST-Ⅳ,测得AST、AST-Ⅳ两种药物各时点血清中AST-Ⅳ浓度前者均是后者3倍以上,且最高浓度前者是后者的3.5倍。结论以AST-Ⅳ血药浓度为指标,AST较AST-Ⅳ更易于吸收。
Objective To establish a solid phase extraction HPLC method for the determination of astragaloside in rat serum. Compare AST and Astragaloside IV (AST-IV) in serum of rats at the same time after intraperitoneal injection in rats. -IV content. Methods Rats were intraperitoneally injected with AST and AST-IV (20 mg/kg equivalent dose according to AST-IV). Rats serum AST-IV was measured at 30, 60, 75, 90, 105, and 120 minutes after administration. And analyze and compare the results of the two drugs. RESULTS: Rats in the same volume of AST-IV were injected intraperitoneally and the AST-IV concentration in the serum of the AST and AST-IV was higher than that of the latter by more than three times at each time point. The highest concentration of the former was 3.5 times that of the latter. . Conclusions Using AST-IV plasma concentration as an index, AST is more easily absorbed than AST-IV.
