目的草杞颗粒对荷瘤小鼠的抗肿瘤作用机制。方法取昆明种小鼠18~22 g,雌雄各半,共50只,接种S180肿瘤细胞在腋窝皮下0.2 ml,然后分组为:阴性组(空白组),24 h后灌胃给水0.5 ml/20 g,1次/d,连续8 d;阳性组,灌胃给5氟尿嘧啶(5-Fu)25 mg/kg;草杞颗粒小剂量组生药2.21 g生药/kg,中剂量组4.43 g/kg,大剂量组8.85 g生药/kg,上述每组均在接种肿瘤后24 h灌胃给药,1次/d,连续8 d,次日称重、剥离瘤块,入10%中性福尔马林固定。用免疫组化法观察促凋亡免疫酶标抗体Bax、Bcl-6、Hsp27的表达。结果草杞颗粒组均见有大片肿瘤细胞坏死、凋亡,小组累计坏死分数在小、中、大剂量组分别为2.28,2.50和2.76,坏死程度随剂量增大呈线性增加关系。促凋亡免疫酶标抗体Bax、Bcl-6、Hsp27较5-Fu组(1.74)有明显的阳性强度表达增加。结论草杞颗粒能促进肿瘤细胞凋亡,抑制S180肿瘤细胞生长周期。
Aim To investigate the anti-tumor effect of Caoba granule on tumor-bearing mice. Methods Kunming mice (18-22 g) were divided into two groups (50 male and 50 female). The S180 tumor cells were subcutaneously injected 0.2 ml into the armpit, then divided into negative group (blank group) and gavage 0.5 ml / 20 g, once a day for 8 days. Compared with 5-Fu group (1.74), Bax, Bcl-6 and Hsp27, the pro-apoptotic immune enzyme-labeled antibodies, showed obvious positive expression. Conclusion Caizhi granule can promote tumor cell apoptosis and inhibit S180 tumor cell growth cycle.
