目的:建立同时测定乌灵胶囊中5-hydroxymellein,5-carboxylmellein,金雀异黄素及5-甲基蜂蜜曲霉素含量的方法。方法:采用高效液相色谱法,Kromasil KR100-5C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.2%磷酸溶液,梯度洗脱;流速1.0 mL·min-1;柱温30℃。结果:5-hydroxymellein在7.86~157.2 ng线性关系良好,平均回收率为101%,RSD1.3%;5-carboxylmellein在9.57~191.4 ng线性关系良好,平均回收率为98.6%,RSD 1.5%;金雀异黄素在28.80~576.0 ng线性关系良好,平均回收率为98.6%,RSD 1.8%;5-甲基蜂蜜曲霉素在21.46~429.2 ng线性关系良好,平均回收率为99.2%,RSD 1.8%。结论:该研究建立的方法可行,重复性好,可用于乌灵胶囊的质量控制。
Objective: To establish a method for simultaneous determination of 5-hydroxymellein, 5-carboxylmellein, genistein and 5-methyl-melphalan in Wuling capsule. Methods: Kromasil KR100-5C18 column (4.6 mm × 250 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile-0.2% phosphoric acid solution with gradient elution. The flow rate was 1.0 mL · min-1. . Results: The linearity of 5-hydroxymellein was 7.86 ~ 157.2 ng, the average recovery was 101% and RSD was 1.3%. The linearity of 5-carboxylmellein was 9.57 ~ 191.4 ng with average recovery of 98.6% and RSD of 1.5% The genistein had a good linear relationship between 28.80 and 576.0 ng, with an average recovery of 98.6% and a RSD of 1.8%. The linear relationship of 5-methylinfevermectin between 21.46 and 429.2 ng was good with an average recovery of 99.2% and RSD 1.8 %. Conclusion: The method established in this study is feasible and reproducible. It can be used for the quality control of Wuling capsule.
