目的:采用HPLC法测定野蔷薇不同部位(根、茎、果实)中齐墩果酸和熊果酸的含量,比较不同产地野蔷薇果实中齐墩果酸和熊果酸含量的差异。方法:采用Zorbax ODS色谱柱(4.6 mm×250 mm×5μm),以甲醇-水(88∶12)为流动相,流速为1.0 m L/min,检测波长为210 nm。结果:齐墩果酸和熊果酸在20 min内较好分离,其质量浓度分别为0.02468~0.2468 mg/m L和0.02523~0.2523 mg/m L时与峰面积线性关系良好,相关系数分别为0.9994和0.9996;平均回收率为分别为99.96%(RSD为0.74%)和100.03%(RSD为0.95%)。结论:该方法操作简便、结果准确、重现性好,可同时测定野蔷薇中齐墩果酸和熊果酸的含量,均在果实中含量最高;库车县、阿图什市的野蔷薇果实中齐墩果酸和熊果酸的含量最多,可作为最适开发利用的固定来源。
OBJECTIVE: To determine the content of oleanolic acid and ursolic acid in different parts (roots, stems and fruits) of Houttuynia cordata Thunb. By HPLC and to compare the content of oleanolic acid and ursolic acid in the hibiscus fruit from different habitats. Methods: Zorbax ODS column (4.6 mm × 250 mm × 5 μm) was used with mobile phase of methanol-water (88:12) as the mobile phase at a flow rate of 1.0 mL / min with a detection wavelength of 210 nm. Results: Oleanolic acid and ursolic acid were separated well within 20 min. The linearity of peak area was good with mass concentrations of 0.02468 ~ 0.2468 mg / m L and 0.02523 ~ 0.2523 mg / m L, respectively. The correlation coefficients were 0.9994 and 0.9996, respectively. The average recoveries were 99.96% (RSD 0.74%) and 100.03% (RSD 0.95%) respectively. Conclusion: The method is simple, accurate, reproducible and can be used for the simultaneous determination of oleanolic acid and ursolic acid in wild rose. The contents of oleanolic acid and ursolic acid are the highest in the fruit. The wild rose in Kuche County and Atushi Oleanolic acid and ursolic acid content of the most fruit can be used as a fixed source of optimal development and utilization.
