目的:研究AngⅡ诱导内皮细胞凋亡作用及阿魏酸钠(SF)的抗凋亡作用和可能机制。方法:将体外培养的猪髂动脉内皮细胞(PIECs)随机分为:对照组、AngⅡ组、SF组、SF+AngⅡ组。倒置显微镜下观察细胞生长状况,MTT法检测PIECs;存活率,流式细胞术(FCM)检测细胞凋亡状况和细胞内活性氧(ROS)水平,比色法法测定上清中MDA含量。结果:与对照组比较,AngⅡ组细胞凋亡率、上清中MDA含量及细胞内ROS水平增加(P<0.01),细胞存活率降低(P<0.01)。SF则降低细胞凋亡率(P<0.05)、上清液MDA含量及细胞内ROS水平(P<0.01),增加细胞存活率(P<0.05),但高浓度的SF有一定的细胞毒性。结论:SF可以抑制Ang诱导的内皮细胞凋亡,该作用与SF降低上清中MDA含量和降低细胞内ROS生成有关。
AIM: To investigate the effect of AngⅡ on endothelial cell apoptosis and the anti-apoptotic effect of sodium ferulate (SF) and its possible mechanism. Methods: Porcine iliac artery endothelial cells (PIECs) were randomly divided into control group, Ang Ⅱ group, SF group and SF + Ang Ⅱ group. The growth of PIECs was observed under inverted microscope. The viability of PIECs was detected by MTT assay. The apoptosis status and intracellular reactive oxygen species (ROS) levels were detected by flow cytometry (FCM). The content of MDA in supernatant was measured by colorimetry. Results: Compared with the control group, the apoptotic rate in AngⅡ group, the content of MDA in supernatant and the intracellular ROS level increased (P <0.01), and the cell survival rate decreased (P <0.01). SF decreased the apoptosis rate (P <0.05), the supernatant MDA content and intracellular ROS level (P <0.01), increased the cell survival rate (P <0.05), but the high concentration of SF had certain cytotoxicity. Conclusion: SF can inhibit the apoptosis of endothelial cells induced by Ang. This effect is related to SF decreasing the content of MDA in the supernatant and decreasing the production of intracellular ROS.
