目的观察川芎嗪对高糖诱导的人视网膜血管内皮细胞(HRCECs)增殖的影响。方法将HRCECs细胞分为对照(生理盐水)组、模型(25 mmol/L葡萄糖)组和川芎嗪低、中、高浓度(50、100、200μmol/L)组,培养48 h后,MTT细胞毒实验检测细胞增殖变化,流式细胞技术检测细胞周期,ELISA法检测血管内皮生长因子(VEGF)表达。结果与对照组比较,高糖对HRCECs细胞增殖、分裂(M)期比例、上清VEGF浓度均具有显著促进作用(P<0.05、0.01);与模型组比较,低、中、高浓度川芎嗪对高糖诱导的HRCECs细胞增殖、分裂(M)期比例、上清VEGF浓度均发挥显著抑制作用(P<0.05、0.01),且呈浓度相关性。结论川芎嗪可能通过抑制高糖诱导的HRCECs细胞VEGF高表达,阻滞细胞周期,发挥抑制HRCECs细胞增殖的作用。
Objective To observe the effect of ligustrazine on the proliferation of human retinal vascular endothelial cells (HRCECs) induced by high glucose. Methods HRCECs were divided into control (saline) group, model group (25 mmol / L glucose) and ligustrazine low, medium and high concentrations (50, 100 and 200μmol / L) Cell proliferation was measured by experiments. Cell cycle was detected by flow cytometry. The expression of vascular endothelial growth factor (VEGF) was detected by ELISA. Results Compared with the control group, high glucose significantly promoted the proliferation, division ratio of M cells and VEGF concentration in supernatant of HRCECs (P <0.05, 0.01). Compared with model group, low, middle and high concentrations of ligustrazine High glucose-induced HRCECs proliferation, division (M) phase ratio, supernatant VEGF concentration were significantly inhibited (P <0.05, 0.01), and showed a concentration-related. Conclusion Tetramethylpyrazine may inhibit the proliferation of HRCECs cells induced by high glucose, which may block the cell cycle and inhibit the proliferation of HRCECs.
