目的:探讨冬虫夏草提取物对结肠癌细胞株HT-29细胞增殖、凋亡的影响及其机制。方法:利用不同浓度冬虫夏草提取物(0.5~6.5 mg/m L)作用HT-29细胞24、48和72 h,采用MTT、平板实验、流式细胞检测和WB检测冬虫夏草提取物对HT-29细胞生长增殖作用、细胞集落数的形成、对细胞凋亡率的影响以及对细胞凋亡相关分子Caspase-3蛋白表达的影响。结果:MTT结果表明,提取物能够显著抑制HT-29的生长增殖,呈一定的量时效关系,半数抑制浓度为3.5 mg/m L;流式细胞仪分析表明,冬虫夏草提取物呈浓度依赖性诱导HT-29细胞凋亡,浓度为6.5 mg/m L时,HT-29凋亡率(12.57±0.16)%;Western blotting表明,HT-29细胞与对照组相比,经不同浓度提取物诱导后,Caspase-3表达逐渐增强。结论:冬虫夏草提取物具有抑制HT-29细胞增殖作用,这可能与增加Caspase-3蛋白表达、诱导细胞调亡有关。
Objective: To investigate the effect and mechanism of Cordyceps sinensis extract on proliferation and apoptosis of colon cancer cell line HT-29. Methods: HT-29 cells were treated with extracts of Cordyceps Sinensis (0.5-6.5 mg / mL) at different concentrations for 24, 48 and 72 h. MTT, plate assay, flow cytometry and WB were used to detect the effects of Cordyceps sinensis extract on HT-29 cells Growth and proliferation, the formation of colony numbers, the effect on the rate of apoptosis and the effect on the expression of Caspase-3 protein. Results: The results of MTT showed that the extract could significantly inhibit the growth and proliferation of HT-29 with a certain amount of time-dependent inhibition and the median inhibitory concentration was 3.5 mg / mL. Flow cytometry analysis showed that Cordyceps sinensis extract was induced in a concentration-dependent manner The apoptosis rate of HT-29 cells in HT-29 cells was 12.57 ± 0.16% at the concentration of 6.5 mg / ml. The results of Western blotting showed that the HT-29 cells treated with extracts of different concentrations , Caspase-3 expression gradually increased. Conclusion: Cordyceps sinensis extract can inhibit the proliferation of HT-29 cells, which may be related to increasing Caspase-3 protein expression and inducing cell apoptosis.
