目的观察卵巢切除大鼠脑内不同部位胆碱乙酰化转移酶(ChAT)阳性细胞表达的变化,了解ChAT改变与轻度认知障碍(MCI)相关的可能机制。方法成年健康雌性Wistar大鼠14只,应用SINGH法建立大鼠卵巢切除模型(模型组),对照组除不切除卵巢外,其余步骤同模型组。常规处死大鼠,取脑组织行ChAT免疫细胞化学染色,观察海马CA1区、皮质区、杏仁复合体区、基底前脑Meynert核等部位ChAT阳性细胞表达变化。结果模型组与对照组比较脑内海马CA1区、大脑皮质区、杏仁复合体区、Meynert核区ChAT阳性细胞数明显减少(t=5.15~16.48,P<0.01)。结论雌激素下降可致大鼠脑内多部位ChAT阳性细胞表达减少,这种病理变化可能与MCI发病有关。
Objective To observe the changes of choline acetyltransferase (ChAT) positive cells in different parts of the ovariectomized rat brain and to find out the possible mechanism of ChAT changes associated with mild cognitive impairment (MCI). Methods Fourteen healthy female Wistar rats were established. The ovariectomized rat model was established by SINGH method. The control group was treated with ovariectomy without any ovariectomy. The rats were sacrificed routinely, and the brain tissue was taken for ChAT immunocytochemical staining to observe the changes of ChAT positive cells in the hippocampal CA1, cortex, almond complex and Meynert nucleus. Results Compared with the control group, the numbers of ChAT positive cells in hippocampus CA1, cerebral cortex, almond complex and Meynert nucleus in model group were significantly decreased (t = 5.15-16.48, P <0.01). Conclusion The decrease of estrogen may decrease the expression of ChAT positive cells in multiple parts of rat brain. This pathological change may be related to the pathogenesis of MCI.
