目的了解急性早幼粒细胞白血病(APL)患者T细胞受体重排删除DNA环(TRECs)的含量,从而了解患者胸腺近期输出na觙veT细胞的水平。方法利用实时定量PCR(TaqMan)方法检测9例APL患者和14例正常人外周血单个核细胞(PBMC)TRECs的水平,并根据外周血中CD3阳性率计算CD3细胞中TRECs水平。结果正常人TRECs拷贝数为(4.10±3.65)/1000PBMC和(6.36±5.28)/1000CD+3细胞,APL患者TRECs拷贝数为(0.13±0.22)/1000PBMC和(0.77±1.60)/1000CD+3细胞,与正常人相比均有显著性差异(P=0.0004,P=0.0005)。结论率先报道了APL患者胸腺近期输出na觙veT细胞功能明显降低的情况。
Objective To understand the content of T cell receptor rearrangement deletion DNA loops (TRECs) in patients with acute promyelocytic leukemia (APL) so as to know the level of na 觙 veT cells in the thymus. Methods The levels of TRECs in peripheral blood mononuclear cells (PBMCs) of 9 APL patients and 14 normal controls were detected by real-time quantitative PCR (TaqMan). The levels of TRECs in CD3 cells were calculated according to the positive rate of CD3 in peripheral blood. RESULTS: The copy number of TRECs was (4.10 ± 3.65) / 1000 PBMC and (6.36 ± 5.28) / 1000 CD + 3 cells in normal controls. The TRECs copy number was (0.13 ± 0.22) / 1000 PBMC and (0.77 ± 1.60) , There were significant differences compared with normal people (P = 0.0004, P = 0.0005). Conclusion The first report of APL patients with thymus output na 觙 veT cell function was significantly reduced.
