目的探讨杨梅素对高糖诱导的血管内皮细胞氧化应激损伤的影响。方法体外培养原代人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs),分为正常对照组(葡萄糖浓度5.56 mmol/L)、甘露醇高渗对照组(5.56 mmol/L葡萄糖+27.8 mmol/L甘露醇)、高糖处理组(葡萄糖浓度33.36 mmol/L)、杨梅素保护组(0.1~10μmo/L杨梅素+33.36 mmol/L葡萄糖),处理48 h后,CCK-8比色法检测细胞活力,Annexin-FITC/PI双染流式细胞术检测细胞凋亡,同时检测细胞内活性氧(ROS)含量,及培养上清液中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性。结果高糖处理HUVECs 48h后,与正常对照组比较,细胞活力显著降低(P<0.05),细胞凋亡率、ROS及MDA含量均显著增加(P<0.05),而SOD和CAT活性显著下降(P<0.05);杨梅素预处理HUVECs后,可显著抑制高糖引起的HUVECs细胞凋亡、ROS和MDA含量的增加以及细胞活力、SOD和CAT活性的下降(P<0.05)。结论杨梅素可减轻高糖诱导的血管内皮细胞氧化应激,抑制细胞凋亡。
Objective To investigate the effect of myricetin on oxidative stress injury induced by high glucose in vascular endothelial cells. Methods Primary human umbilical vein endothelial cells (HUVECs) were cultured in vitro and divided into normal control group (glucose concentration 5.56 mmol / L), mannitol hypertonic control group (5.56 mmol / L glucose + 27.8 mmol / L mannitol), high glucose (33.36 mmol / L glucose) and myricetin (0.1-10 μmo / L myricetin +33.36 mmol / L glucose) Cell viability and apoptosis were detected by Annexin-FITC / PI double staining flow cytometry. The level of intracellular reactive oxygen species (ROS) and the contents of malondialdehyde (MDA), superoxide dismutase SOD) and catalase (CAT) activity. Results Compared with the normal control group, the viability of HUVECs was significantly decreased (P <0.05), the apoptotic rate, the content of ROS and MDA significantly increased (P <0.05) and the activities of SOD and CAT decreased significantly P <0.05). Pretreatment with myricetin significantly inhibited the apoptosis of HUVECs induced by high glucose, the increase of ROS and MDA content, the decrease of cell viability and the activity of SOD and CAT (P <0.05). Conclusion Myricetin can reduce high glucose-induced oxidative stress in vascular endothelial cells and inhibit apoptosis.
