目的:探讨癌胚抗原黏附分子1对胃癌细胞株AGS生物学特性的影响。方法:将CEACAM1 si RNA转染人胃癌细胞株AGS,再用western-blot检测CEACAM1的表达;流式细胞术、Transwell实验和裸鼠负荷瘤实验分析转染后的AGS细胞的生物学特性变化。结果:转染CEACAM1 si RNA后的AGS细胞几乎不表达CEACAM1。CEACAM1沉默基因对AGS细胞增殖、凋亡和细胞周期无影响。转染CEACAM1 si RNA后的AGS细胞侵袭能力明显降低,荷瘤裸小鼠肿瘤的生长受到抑制。结论:沉默CEACAM1基因的表达能够明显抑制AGS细胞的转移以及活体成瘤性,为胃癌的基因治疗提供理论依据。
Objective: To investigate the effect of CEA on the biological characteristics of gastric cancer cell line AGS. Methods: CEACAM1 si RNA was transfected into human gastric cancer cell line AGS, and the expression of CEACAM1 was detected by western-blot. The biological characteristics of AGS cells were analyzed by flow cytometry, Transwell assay and nude mice bearing tumor. Results: AGS cells transfected with CEACAM1 si RNA seldom expressed CEACAM1. CEACAM1 silencing gene had no effect on AGS cell proliferation, apoptosis and cell cycle. The invasion ability of AGS cells transfected with CEACAM1 si RNA was significantly reduced, and the growth of tumor-bearing nude mice was inhibited. Conclusion: The silencing of CEACAM1 gene expression can significantly inhibit AGS cell metastasis and tumorigenicity in vivo, providing a theoretical basis for gene therapy of gastric cancer.
