目的研究近红外量子点(QD800)对人舌鳞癌细胞(Tca8113细胞)的生长、成瘤、增殖及凋亡的影响。方法 (1)QD800对Tca8113细胞标记后6 h、12 h、24 h、3 d、5 d、7 d用流式细胞仪检测细胞的标记率及平均荧光强度。(2)用浓度为10 nmol/L的QD800标记的Tca8113细胞及未标记的Tca8113细胞行裸鼠皮下成瘤,观察QD800对Tca8113细胞成瘤能力的影响(。3)用流式细胞仪检测QD800对Tca8113细胞增殖及凋亡的影响。结果 (1)QD800对Tca8113细胞标记后6 h、12 h、24 h、3 d、5 d、7 d用流式细胞仪检测细胞的标记率分别为92.30%、77.07%、67.36%、49.59%、15.25%、8.37%,平均荧光强度分别为211.23、187.63、113.08、100.47、85.86、47.28。(2)Tca8113/QD800组和Tca8113组在皮下接种24 d后肿瘤的平均重量分别为(1.22±0.12)g、(1.25±0.16)g。(3)Tca8113/QD800和Tca8113体内成瘤24 d后细胞的平均增殖指数为(48.21±1.13)%、(47.25±1.27)%,平均凋亡指数为(11.33±0.65)%、(10.66±0.57)%。结论用近红外量子点QD800标记Tca8113细胞后不影响其生长、成瘤、增殖及凋亡能力。
Objective To study the effect of QD800 on the growth, tumorigenesis, proliferation and apoptosis of human tongue squamous cell carcinoma (Tca8113) cells. Methods (1) QD800 was used to detect the labeling rate and average fluorescence intensity of Tca8113 cells after 6 h, 12 h, 24 h, 3 d, 5 d and 7 d of flow cytometry. (2) Tca8113 cells labeled with QD800 and untagged Tca8113 cells at a concentration of 10 nmol / L were subcutaneously injected into nude mice to observe the effect of QD800 on tumorigenicity of Tca8113 cells (.3) Flow cytometry was used to detect QD800 Effects on Tca8113 Cell Proliferation and Apoptosis. Results (1) The labeling rates of cells detected by flow cytometry at 6 h, 12 h, 24 h, 3 d, 5 d, and 7 d after QD800 labeling were 92.30%, 77.07%, 67.36%, 49.59% , 15.25% and 8.37% respectively. The average fluorescence intensity was 211.23, 187.63, 113.08, 100.47, 85.86 and 47.28 respectively. (2) The mean tumor weights of Tca8113 / QD800 group and Tca8113 group after subcutaneous inoculation for 24 d were (1.22 ± 0.12) g and (1.25 ± 0.16) g, respectively. (3) The average proliferation index of Tca8113 / QD800 and Tca8113 in vivo after 24-day tumor growth was (48.21 ± 1.13)% and (47.25 ± 1.27)% respectively, and the average apoptotic index was 11.33 ± 0.65% and 10.66 ± 0.57 respectively )%. Conclusion The labeling of Tca8113 cells with QD800 near NIR did not affect the growth, tumorigenesis, proliferation and apoptosis of Tca8113 cells.
