目的:建立高效液相色谱法(HPLC)测定柿叶中齐墩果酸含量的方法。方法:采用高效液相色谱法测定柿叶中齐墩果酸的含量,色谱柱为UitimateC18(4.6mm×250mm×5μm),流动相为甲醇-0.5%乙酸铵(85∶15),流速1.0mL·min-1,检测波长210nm,柱温25℃。结果:齐墩果酸含量在0.050~0.500μg/mL之间与峰面积呈良好的线性关系,回归方程Y=4680.2X+7.4412,r=0.9999,平均回收率为99.5%,RSD为0.507%(n=6)。结论:该方法简便,灵敏度高,重复性好,可用于中药柿叶的质量评价。
Objective: To establish a method for the determination of oleanolic acid in persimmon leaves by high performance liquid chromatography (HPLC). Methods: The content of oleanolic acid in persimmon leaves was determined by high performance liquid chromatography (HPLC) with Uitimate C18 column (4.6 mm × 250 mm × 5 μm), methanol-0.5% ammonium acetate (85:15) Min-1, detection wavelength 210nm, column temperature 25 ℃. Results: The content of oleanolic acid in the range of 0.050-0.500 μg / mL showed a good linear relationship with the peak area. The regression equation was Y = 4680.2X + 7.4412, r = 0.9999. The average recovery was 99.5% and the RSD was 0.507% n = 6). Conclusion: The method is simple, sensitive and reproducible. It can be used to evaluate the quality of Chinese persimmon leaves.
