目的观察温经汤对寒凝血瘀模型大鼠卵巢血红素氧合酶1(HO-1)、血红素氧合酶2(HO-2)的影响,探讨温经汤治疗寒凝血瘀型妇科疾病的作用机制。方法按随机数字表法将40只SD雌性大鼠分为正常组、模型组、温经汤低剂量组(低剂量组)、温经汤高剂量组(高剂量组),每组10只。低剂量组、高剂量组于造模的第1天开始,分别按14.66、29.32g/(kg.d)-1给予温经汤灌胃,每日1次,连用2周。其余两组给予同体积的蒸馏水。采用血红蛋白结合及联二亚硫酸盐还原法测定血浆一氧化碳(CO)活性;采用免疫组织化学SP法检测卵巢颗粒细胞和黄体细胞HO-1、HO-2蛋白表达;采用原位杂交法检测卵巢颗粒细胞和黄体细胞HO-1、HO-2 mRNA的表达。结果高剂量组、低剂量组大鼠血浆碳氧血红蛋白(COHb)活性较模型组明显增强(P<0.05或P<0.01);卵巢颗粒细胞、黄体细胞HO-1、HO-2蛋白表达,HO-1 mRNA、HO-2 mRNA表达加强(P<0.05或P<0.01)。结论温经汤治疗寒凝血瘀型妇科疾病的作用可能与调节HO-1、HO-2水平有关。

Conclusion The effect of Wenjing Decoction on the treatment of gynecological diseases with cold coagulation and blood stasis may be related to the regulation of HO-1 and HO-2 levels.
