采用大孔树脂吸附法纯化姜油树脂,运用静态吸附与解吸试验筛选到X-5树脂对姜辣素具有较好的吸附和解吸性能。通过单因素和正交试验探讨了样品浓度、吸附速率、乙醇解吸液浓度、解吸速率以及解吸用量等对X-5树脂纯化的影响。结果表明:2.24 mg/mL浓度样品以1 mL/min速率吸附,吸附率最高可达87.69%,2倍柱体积的75%乙醇以0.5 mL/min速率解吸,纯化后姜辣素纯度达到32.78%。
The macroporous resin was used to purify ginger oleoresin. X-5 resin was screened by static adsorption and desorption test, which showed good adsorption and desorption properties to gingerol. The effects of sample concentration, adsorption rate, concentration of ethanol desorption solution, desorption rate and desorption amount on the purification of X-5 resin were discussed by single factor and orthogonal experiments. The results showed that when the concentration of 2.24 mg / mL was 1 mL / min, the adsorption rate was up to 87.69%. After 2 column volumes of 75% ethanol was desorbed at 0.5 mL / min, the purity of the purified gingerol reached 32.78% .
