目的建立HPLC法测定复方鱼腥草合剂中绿原酸、黄芩苷和汉黄芩素的含量,为中药复方中多成分检测寻求可行的方法。方法采用Shim-pack VP-ODS(250 mm×4.6 mm,5μm)色谱柱;流动相为乙腈-甲醇-0.2%磷酸溶液,梯度洗脱;流速:0.8 ml·min-1,检测波长:275 nm。绿原酸、黄芩苷和汉黄芩素与其相邻杂质峰能完全分离。结果绿原酸、黄芩苷和汉黄芩素的线性范围分别为1.96~39.1μg·ml-1(r=0.9999)、6.25~124.9μg·ml-1(r=0.9999)、0.26~5.24μg·ml-1(r=0.9998)。方法回收率均>98%。结论本方法简便、准确、重复性好,能排除其他成分的干扰,可用于该制剂的质量控制。
OBJECTIVE To establish a HPLC method for the determination of chlorogenic acid, baicalin and wogonin in compound Houttuynia mixture, and to find a feasible method for the multi-component determination of traditional Chinese medicine compound prescriptions. Methods The chromatographic column was Shim-pack VP-ODS (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-methanol-0.2% phosphoric acid solution with gradient elution. The flow rate was 0.8 ml · min- . Chlorogenic acid, baicalin and wogonin with its adjacent impurity peaks can be completely separated. Results The linear ranges of chlorogenic acid, baicalin and wogonin were 1.96-39.1μg · ml-1 (r = 0.9999), 6.25-124.9μg · ml-1 (r = 0.9999), 0.26-5.24μg · ml -1 (r = 0.9998). The recovery rates were> 98%. Conclusion The method is simple, accurate, reproducible and can eliminate the interference of other components and can be used for the quality control of the preparation.
