目的研究补阳还五汤的有效成分黄芪甲苷、阿魏酸、芍药苷、红花黄色素、川芎嗪对阿尔茨海默症(AD)模型的影响。方法采用MTT法检测不同浓度Aβ1-40对大鼠的微血管内皮细胞(BMVEC)活性的影响;并检测补阳还五汤中各有效成分对AD大鼠模型BMVEC增殖的影响。结果补阳还五汤的有效成分对AD模型有作用,各剂量黄芪甲苷、阿魏酸、芍药苷、红花黄色素、川芎嗪均可降低AD模型BMVEC增殖率(P<0.05),且对细胞增殖的抑制作用随浓度的增加而增强,以黄芪甲苷的作用最为显著。结论补阳还五汤的有效成分可显著抑制AD模型BMVEC增殖,为补阳还五汤对AD的治疗奠定实验基础。
Objective To study the effects of Astragaloside, ferulic acid, paeoniflorin, safflower yellow and tetramethylpyrazine on the Alzheimer’s disease (AD) model in Buyang Huanwu Decoction. Methods The effects of different concentrations of Aβ1-40 on the microvessel endothelial cells (BMVEC) activity in rats were measured by MTT assay. The effects of various active ingredients in Buyang Huanwu Decoction on the proliferation of BMVECs were observed. Results The active ingredients of Buyang Huanwu Decoction had an effect on AD model. Astragaloside, ferulic acid, paeoniflorin, safflower yellow and tetramethylpyrazine could all reduce the proliferation rate of BMVEC in AD model (P <0.05) The inhibitory effect on cell proliferation increased with increasing concentration, the most significant role of Astragaloside. Conclusion The active ingredients of Buyang Huanwu Decoction can significantly inhibit the proliferation of BMVEC in AD model and lay the experimental foundation for the treatment of AD with Buyang Huanwu Decoction.
