目的观察不同浓度复方奥硝唑-甲磺酸培氟沙星缓释制剂对人牙周膜细胞(human periodontal ligament cells,HPDLC)生长及ki-67表达的影响。方法组织块法培养原代HPDLC,MTT法检测HPDLC在复方奥硝唑-甲磺酸培氟沙星缓释制剂的不同浓度(0、1.25、2.5、5、10、20 g/L)下的光密度值,检测细胞的相对增殖率;采用流式细胞仪检测细胞周期;免疫荧光法检测ki-67的阳性表达率。结果在1.25、2.5 g/L的浓度下对HPDLC的生长和ki-67的表达与对照组比较差异均无统计学意义(P>0.05);而5、10 g/L浓度组的光密度值有所增大,细胞增殖指数提高,ki-67的阳性表达率也有所增加,与对照组比较差异有统计学意义(P<0.05);高浓度药物20 g/L对HPDLC的生长有一定的抑制作用。结论 5、10 g/L的复方奥硝唑-甲磺酸培氟沙星缓释制剂对体外培养的HPDLC的生长增殖有一定促进作用。
