目的评价复方透骨草溶液的抑菌和抗炎作用,初步探索其抗炎的作用机制,为临床应用提供实验依据。方法采用体外抑菌实验,观察复方透骨草溶液对常见皮肤癣菌和酵母菌的抑制作用;采用二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足趾肿胀的模型,观察复方透骨草溶液和地塞米松磷酸钠注射液外用涂抹的抗炎作用,并检测大鼠足趾致炎4 h后组织中的白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的水平。结果体外抑菌实验表明,复方透骨草溶液对皮肤癣菌和酵母菌的最小抑菌浓度(MIC)分别为原液浓度的12.5%和25.0%;其能降低二甲苯致小鼠耳廓的肿胀度,高剂量给药组与阴性对照组比较差异有统计学意义(P<0.01);对角叉菜胶致大鼠足趾肿胀有明显的减轻作用,给药组与阴性对照组比较差异均有统计学意义(P<0.01);能显著降低大鼠足趾肿胀损伤后组织中致炎因子IL-1β的表达,给药组与阴性对照组比较差异均有统计学意义(P<0.05,P<0.01)。结论复方透骨草溶液具有显著的抑菌和抗炎作用,并通过降低组织中的炎症因子IL-1β的水平,从而抑制炎症发展的过程。
OBJECTIVE To evaluate the antimicrobial and anti-inflammatory effects of the compound Hupengcao solution and to explore its anti-inflammatory mechanism to provide experimental evidence for clinical application. Methods In vitro antibacterial experiment was used to observe the inhibitory effect of the compound Huachagou solution on common dermatophytes and yeast. The xylene-induced mouse ear swelling and carrageenan-induced rat toe swelling were observed. Anti-inflammatory effect of topical solution of dexamethasone and sodium dexamethasone sodium phosphate injection and detection of interleukin-1β (IL-1β) and tumor necrosis factor-α -α) levels. Results The antibacterial activity in vitro showed that the minimum inhibitory concentration (MIC) of the solution of the extract of Fructus Fortunei to Trichophyton mentagrophytes and yeast was 12.5% and 25.0% of that of the original solution, respectively. It could reduce the swelling of xylene-induced auricle in mice (P <0.01); carrageenan-induced rat toe swelling significantly reduce the role of drug-administered group and the negative control group were significantly different (P <0.01). It could significantly reduce the expression of IL-1β in the tissue after the toe swelling in rats. There was significant difference between the drug group and the negative control group (P <0.05, P <0.01). CONCLUSION The compound Hupengcao solution has remarkable antibacterial and anti-inflammatory effects and can inhibit the process of inflammation by reducing the level of IL-1β, an inflammatory factor in the tissue.
