小鼠脑梗死后血浆ET-1,TXA_2,PGI_2的变化及尼莫同的影响

目的 观察小鼠脑梗死后血浆中ET-1,TXA_2,PGI_2的变化以及尼莫同的影响。方法经右侧颈总动脉将尼龙单丝线栓至大脑中动脉造成永久性缺血模型。经尾静脉及腹腔注射尼莫同。经左心室采血,放免法测定血浆ET-1,TXA_2,PGI_2的含量。结果单纯缺血组ET-1,TXA_2明显升高,PGI_2明显降低。术后第1d、2d、3d、4d、5d开始使用尼莫同均可降低ET-1,TXA_2,而升高PGI_2。结论 脑缺血后不同时间开始使用尼莫同均可降低ET-1,TXA_2,而升高PGI_2。 Objective To observe the changes of ET-1, TXA 2 and PGI 2 in plasma and the influence of Nimodipine after cerebral infarction in mice. Methods A permanent ischemic model was established by tethering nylon monofilaments to the middle cerebral artery via the right common carotid artery. Nemo tail through the tail vein and intraperitoneal injection. Left ventricular blood collection, radioimmunoassay determination of plasma ET-1, TXA_2, PGI_2 content. Results The levels of ET-1 and TXA2 in ischemia group were significantly increased, and PGI_2 was significantly decreased. After the first day, 2d, 3d, 4d, 5d, the use of nimotone both decreased ET-1 and TXA_2 and increased PGI_2. Conclusions Nimodipine can decrease ET-1 and TXA2 and increase PGI_2 at different time points after cerebral ischemia.
