目的探讨丹红注射液(丹参和红花)对心梗(MI)模型大鼠缺血心肌血管新生的影响及其作用机制。方法利用Sprague-Dawley(SD)大鼠左前降支冠状动脉结扎法造成急性心梗模型,实验分为假手术组,模型组,丹红注射液低、中、高剂量组(0.5、1、2 m L/kg),静脉给药7 d后处死,检测心梗面积,心肌损伤血清学指标,微血管面密度,血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)、基质细胞衍生因子-l(stromal cell-derived factor-1,SDF-l)及其受体CXCR4的表达。结果丹红注射液可显著减小大鼠心肌梗死面积,降低LDH、CK-MB、c Tn I血清水平,增加微血管面密度,上调VEGF、SDF-l、CXCR4的表达。结论丹红注射液具有缺血心肌的保护作用,其机制可能与VEGF的高表达及SDF-l/CXCR4生物轴有关。
Objective To investigate the effect of Danhong injection (Salvia miltiorrhiza and safflower) on angiogenesis in ischemic myocardium of myocardial infarction (MI) model rats and its mechanism. Methods The model of acute myocardial infarction was induced by ligation of the left anterior descending coronary artery in Sprague-Dawley (SD) rats. The experimental groups were divided into sham operation group, model group, Danhong injection group, low, medium and high dose groups The rats were sacrificed 7 days after intravenous administration. The myocardial infarction area, myocardial injury, serum microvascular density, vascular endothelial growth factor (VEGF), stromal cell-derived factor-1 ( stromal cell-derived factor-1, SDF-1) and its receptor CXCR4. Results Danhong injection can significantly reduce myocardial infarct size, reduce serum levels of LDH, CK-MB and cTnI, increase microvessel density, and up-regulate the expression of VEGF, SDF-1 and CXCR4. Conclusion Danhong injection has the protective effect of ischemic myocardium, and its mechanism may be related to the high expression of VEGF and the biological axis of SDF-1 / CXCR4.
