目的:探讨激活素A(activin A)对子宫内膜异位症(endometriosis,EMs)患者在位子宫内膜间质细胞(endometrial stromal cells,ESCs)活性的调节作用。方法:分别用RT-PCR和Real-timePCR的方法检测5例正常、45例EMs在位、异位组织中激活素AβA亚基和芳香化酶CYP19A1mRNA的表达。分离培养5例正常、5例EMs在位、异位组织ESCs,用不同剂量的激活素A刺激EMs在位ESCs不同时间后,应用免疫电化学发光法检测ESCs分泌雌二醇(E2)的水平,MTT检测ESCs活性,流式细胞仪检测ESCs凋亡和细胞周期。结果:激活素AβA亚基在EMs异位ESCs的表达明显高于正常对照ESCs和EMs在位ESCs。经25 ng/ml激活素A刺激24 h后,与正常对照组相比,在位ESCs的E2分泌和芳香化酶CYP19A1mRNA的表达均显著升高。但激活素A未引起ESCs增殖活性的显著变化,对细胞凋亡和细胞周期亦无影响。结论:激活素A可能通过促进芳香化酶表达而增加ESCs的E2分泌,这可能是EMs异位灶存活和病理发生的机制之一。
Objective: To investigate the regulatory effect of activin A on the activity of endometrial stromal cells (ESCs) in patients with endometriosis (EMs). Methods: The expressions of activin AβA subunit and aromatase CYP19A1 mRNA in 5 eutopic and ectopic EMs were detected by RT-PCR and Real-time PCR respectively. Five normal and five EMs ESCs were isolated and cultured, and ESCs were stimulated with different doses of activin-A for different time. The levels of estradiol (E2) secreted by ESCs were detected by immunoelectrochemiluminescence The activity of ESCs was detected by MTT and the apoptosis and cell cycle of ESCs were detected by flow cytometry. Results: The expression of activin AβA subunit in ectopic ESCs of EMs was significantly higher than that of normal control ESCs and ESCs in EMs. After stimulation with 25 ng / ml activin A for 24 h, the E2 secretion and the expression of aromatase CYP19A1 mRNA in ESCs were significantly increased compared with the normal control group. However, activin A did not cause significant changes in proliferative activity of ESCs, and had no effect on apoptosis and cell cycle. Conclusion: Activin A may increase E2 secretion of ESCs by promoting the expression of aromatase, which may be one of the mechanisms of ectopic focus survival and pathology.
