目的 探讨姜黄素对人结肠癌细胞Lovo体外生长及其细胞周期的影响。方法 MTT法检测肿瘤细胞活性 ,流式细胞仪进行细胞周期时相分析。结果 MTT结果表明姜黄素对Lovo细胞具有细胞毒作用。 2 0 μM、40 μM浓度的姜黄素对细胞的抑制率分别为 41 9%、78.1% ,5~ 10 μM浓度抑制作用较小。姜黄素处理 2 4h ,分裂相细胞的比例可达 8~ 10 %。FCM结果显示姜黄素处理 2 4h时 ,主要使细胞阻滞于S ,G2 +M期 ,使此期的细胞比例增加 ,而G0 / 1期的细胞比例减少。处理 48h后 ,G2 +M期的细胞比例反而降低 ,部分可能由于G2 +M期细胞并发凋亡所致。结论 姜黄素可抑制结肠癌Lovo细胞的生长。其机制可能与干扰细胞周期有关
Objective To investigate the effect of curcumin on growth and cell cycle of human colon cancer cell line Lovo in vitro. Methods The activity of tumor cells was measured by MTT assay, and the cell cycle was analyzed by flow cytometry. Results MTT results showed that curcumin had cytotoxic effects on Lovo cells. The inhibitory rates of curcumin at concentrations of 20 μM and 40 μM were 41.9% and 78.1%, respectively. The inhibitory effect of 5 to 10 μM concentration was small. After curcumin treatment for 24 hours, the proportion of dividing cells can reach 8 to 10%. The results of FCM showed that when curcumin was treated for 24 hours, the cells were mainly arrested in S and G2 + M phases, which increased the proportion of cells in this phase, while the percentage of cells in G0 / 1 phase decreased. After 48 hours of treatment, the proportion of cells in the G2 + M phase decreased, which may be due to the apoptosis of cells in the G2 + M phase. Conclusion Curcumin can inhibit the growth of colon cancer Lovo cells. Its mechanism may be related to interference with cell cycle
