为探讨急性妊娠脂肪肝(AFLP)患者肝组织中肝细胞凋亡与肝细胞再生的状况,本文采用了免疫组化技术,以抗-Fas多克隆抗体和抗-增殖细胞核抗原(PCNA)单克隆抗体对石蜡包埋的26例活检或尸解肝组织中Fas抗原及PCNA的表达和分布进行了研究。26例肝组织Fas抗原检测均为阳性(100%),Fas抗原强烈地表达于肝细胞浆内。PCNA的检出率为23.1%(6/26),阳性物质位于核内呈核型表达,阳性细胞少而稀疏地分布于肝小叶内。结果提示:AFLP的发病机理可能与Fas系统的免疫调控密切相关,肝细胞的大量凋亡以及肝细胞再生能力的低下致使AFLP患者预后极差
In order to investigate the status of hepatocyte apoptosis and hepatocyte regeneration in liver of patients with acute pregnancy fatty liver (AFLP), immunohistochemistry was used to detect the expression of anti-Fas polyclonal antibody and anti-proliferating cell nuclear antigen (PCNA) monoclonal antibody Antibodies The expression and distribution of Fas antigen and PCNA in 26 paraffin-embedded biopsies or autopsy liver tissues were studied. Fas antigen was detected in all 26 cases (100%). Fas antigen was strongly expressed in the cytoplasm of liver cells. The detection rate of PCNA was 23.1% (6/26). The positive substance was located in the nucleus with karyotype expression. The positive cells were sparsely distributed in the hepatic lobule. The results suggest that the pathogenesis of AFLP may be closely related to the immune regulation of Fas system. The large number of apoptotic hepatocytes and the low regenerative capacity of hepatocytes lead to poor prognosis in patients with AFLP
