目的:采用中心组合(Box-Behnken)设计优选南方红豆杉[Taxus mairei(Lemee et Levl.)S.Y.Hu ex Liu]枝条中多糖的微波提取工艺。方法:在单因素实验基础上,采用Box-Behnken设计对影响南方红豆杉枝条中多糖提取率的主要因素(料液比、提取时间、提取次数)进行优化,建立影响因素与响应值之间的数学模型,确定最佳超声提取工艺条件。结果:南方红豆杉枝条中多糖最佳超声提取工艺为:料液比21.48(V/W),提取时间13.78 min,提取次数3次,微波频率为1887MHz。结论:采用响应面法对南方红豆杉中多糖的提取条件进行优化,该方法合理可靠,能为生产工艺提供理论依据。
OBJECTIVE: To optimize the microwave extraction process of polysaccharides in Taxus mairei (Lemee et Levl.) S.Y.Hu ex Liu] shoots using the Box-Behnken design. Methods: Based on the single-factor experiment, the main factors influencing the extraction rate of polysaccharide from the branches of Taxus chinensis var. Mairei were determined by the Box-Behnken design (the ratio of solid to liquid, the extraction time and the number of extraction), and the relationship between the influencing factors and the response Mathematical model to determine the best ultrasonic extraction process conditions. Results: The optimal ultrasonic extraction process of polysaccharides from the branches of Taxus chinensis was: the ratio of material to liquid was 21.48 (V / W), the extraction time was 13.78 min, the number of extraction was 3 and the microwave frequency was 1887 MHz. Conclusion: The extraction conditions of polysaccharides in Taxus cuspidata were optimized by response surface methodology. The method is reasonable and reliable and can provide theoretical basis for the production process.
