目的:研究从大豆中分离纯化多烯磷脂酰胆碱的最佳提纯工艺。方法:以浓缩大豆磷脂为原料,采用丙酮和乙醇为溶剂,经丙酮脱油、乙醇抽提、减压蒸馏浓缩、层析柱分离制备高纯度多烯磷脂酰胆碱的提纯工艺。结果:在40℃采用丙酮萃取30min,95%乙醇20~30℃抽提2h,再经氧化铝层析柱层析,多烯磷脂酰胆碱含量可富集到98%以上。结论:此方法提取条件简便,操作简单,提取纯度比较高。
Objective: To study the best purification technology of polyene phosphatidylcholine isolated from soybean. Methods: The purified soybean phosphatidylcholine was used as raw material, acetone and ethanol were used as solvent, and the product was purified by acetone de-oiling, ethanol extraction and vacuum distillation. The purification process of high-purity polyene phosphatidylcholine was achieved by chromatographic separation. Results: Extraction with acetone at 40 ° C for 30 min and 95% ethanol extraction at 20-30 ° C for 2 h followed by alumina column chromatography indicated that polyene phosphatidylcholine content could be enriched to over 98%. Conclusion: This method has the advantages of simple extraction conditions, simple operation and high extraction purity.
