目的首次以藁本内酯纯度和转移率为主要考察指标,探讨应用超临界CO2萃取法从当归中提取总苯酞的工艺。方法采用单因素和L9(3)4正交设计进行试验,考察药材粒度、萃取压力、萃取时间、萃取温度等因素对藁本内酯纯度和转移率的影响,优化最佳提取工艺。采用高效液相色谱法分析,色谱条件为Accurasil C18柱(250mm×4.6mm,5μm),流动相为甲醇-0.05%磷酸水溶液,柱温30℃,检测波长320nm,体积流量1.0mL/min。结果超临界CO2萃取法的最佳工艺条件为:萃取压力10MPa,萃取温度50℃,萃取时间20min。结论超临界CO2流体工艺萃取的当归总苯酞纯度和转移率可达80%以上。
Objective To study the purity and metastasis rate of ligustilide for the first time and to explore the technology of extracting total phthalide from Angelica sinensis by supercritical CO2 extraction. Methods The single factor and orthogonal design of L9 (3) 4 were used to test the effect of drug particle size, extraction pressure, extraction time and extraction temperature on the purity and transfer rate of ligustilide, and the optimum extraction process was optimized. The chromatographic conditions were Accurasil C18 column (250mm × 4.6mm, 5μm), mobile phase consisted of methanol-0.05% phosphoric acid solution, the column temperature was 30 ℃, the detection wavelength was 320nm, and the volume flow rate was 1.0mL / min using high performance liquid chromatography. Results The optimum conditions of supercritical CO2 extraction were: extraction pressure 10MPa, extraction temperature 50 ℃, extraction time 20min. Conclusion The purity and transfer rate of the total phthalide in Angelica sinensis extracted by supercritical CO2 fluid extraction can reach more than 80%.
