目的:探讨淋巴细胞功能相关抗原-1(LFA-1)对活动性狼疮肾炎(LN)外周血单个核细胞(PBMC)白介素 10(IL-10)的调节作用。方法:利用半定量 RT-PCR技术观察 LFA- 1共刺激对活动性 LN患者 PBMC IL-10 mRNA表达的影响。结果:单独抗CD3抗体(30 ng/ml)能诱导LN 患者PBMC IL-10 mRNA轻微表达(0.43+0.03vs 0.55±0.44,P<0.05)而在抗CD3抗体(30 ng/ml)存在的情况下,抗LFA-1抗体(2 μg/ml,相当于LFA-1配体)共刺激时明显增加了PBMC IL—10 mRNA表达(1.31±0.08vs 0.55+0,04,P<0.01)。LFA-1中和抗体明显抑制了这种共刺激诱导的IL—10 mRNA表达(1.31±0.08vs 0.64 ±0.03,P<0.01)。抗LFA-1抗体单独刺激时并不引起 IL-10 mRNA表达变化(P>0.05)。结论:LFA-1作为共刺激分子诱导 IL- 10 mRNA表达可能是其参与 LN发病的机制之一。
Objective: To investigate the regulatory effect of LFA-1 on interleukin-10 (IL-10) in peripheral blood mononuclear cells (PBMC) of patients with active lupus nephritis (LN). Methods: The effects of LFA-1 co-stimulation on IL-10 mRNA expression in PBMC of patients with active LN were observed by semi-quantitative RT-PCR. Results: The anti-CD3 antibody (30 ng / ml) induced a slight expression of IL-10 mRNA in PBMC of LN patients (0.43 + 0.03 vs 0.55 ± 0.44, P <0.05) IL-10 mRNA expression was significantly increased in PBMCs (1.31 ± 0.08 vs. 0) when co-stimulated with anti-LFA-1 antibody (2 μg / ml, equivalent to LFA-1 ligand) 55 + 0,04, P <0.01). Neutralizing antibody to LFA-1 significantly inhibited this costimulation-induced IL-10 mRNA expression (1.31 ± 0.08 vs. 0.64 ± 0.03, P <0.01). Anti-LFA-1 antibody alone did not induce changes in IL-10 mRNA expression (P> 0.05). Conclusion: LFA-1 as costimulatory molecule induces IL-10 mRNA expression may be one of the mechanisms involved in the pathogenesis of LN.
