目的:探讨基因沉默MTA1对宫颈癌CaSki细胞株生物学效应的影响。方法:构建针对MTA1基因的siRNA,体外转染人宫颈癌CaSki细胞株,通过RT-PCR检测其对CaSki细胞MTA1 mRNA表达的影响,并检测转染前后细胞增殖、生长及细胞周期改变情况。结果:瞬时转染MTA1-siRNA入CaSki细胞,可见细胞中MTA1 mRNA表达降低,转染后细胞生长减慢,克隆形成及划痕愈合能力下降,但对细胞周期的改变不明显。随着干扰浓度的增加,时间的延长,siRNA对MTA1 mRNA表达的抑制作用逐渐增强。结论:基因沉默MTA1能使宫颈癌细胞生长减慢,细胞迁移能力下降,一定程度上逆转了宫颈癌细胞的恶性表型。
Objective: To investigate the effect of gene silencing MTA1 on the biological effects of cervical cancer CaSki cell line. Methods: siRNA targeting MTA1 gene was constructed and transfected into human cervical cancer cell line CaSki in vitro. The expression of MTA1 mRNA in CaSki cells was detected by RT-PCR. The proliferation, growth and cell cycle changes were detected before and after transfection. Results: Transient transfection of MTA1-siRNA into CaSki cells showed that the expression of MTA1 mRNA was down-regulated and the cell growth was slowed down after transfection. The ability of cloning and scar healing decreased but the cell cycle was insignificant. With the increase of interference concentration and time, the inhibitory effect of siRNA on MTA1 mRNA expression gradually increased. Conclusion: Gene silencing of MTA1 can slow down the growth of cervical cancer cells and decrease the ability of cell migration, and reverse the malignant phenotype of cervical cancer cells to a certain extent.
