目的研究MTSS1基因在膀胱癌中组织的表达及其对膀胱癌EJ138细胞增殖和凋亡的影响。方法荧光定量PCR检测41例膀胱癌及相应癌旁正常组织中MTSS1基因的表达。将MTSS1基因特异性的siRNA转染EJ138细胞。转染后,Western Blot检测转染后MTSS1蛋白的表达,MTT法测定细胞生长抑制率,流式细胞法检测细胞凋亡率。结果与癌旁正常组织相比,MTSS1 mRNA在膀胱癌组织中表达明显下调。RNA干扰能够显著抑制EJ138细胞中MTSS1蛋白的表达;MTSS1基因表达降低后,EJ138细胞的生长抑制率明显降低,细胞增殖活力明显增加,而且凋亡率明显降低。结论 MTSS1基因在膀胱癌低表达,促进膀胱癌细胞增殖并抑制细胞凋亡。
Objective To investigate the expression of MTSS1 in bladder cancer tissue and its effect on the proliferation and apoptosis of bladder cancer EJ138 cells. Methods The expression of MTSS1 in 41 cases of bladder cancer and corresponding normal tissues was detected by fluorescence quantitative PCR. The MTSS1 gene-specific siRNA was transfected into EJ138 cells. After transfection, the expression of MTSS1 protein was detected by Western Blot. The cell growth inhibition rate was determined by MTT assay and the apoptosis rate by flow cytometry. Results The expression of MTSS1 mRNA was significantly down-regulated in bladder cancer as compared with the adjacent normal tissues. RNA interference could significantly inhibit the expression of MTSS1 in EJ138 cells. After the expression of MTSS1 was down-regulated, the growth inhibition rate of EJ138 cells was significantly decreased, the cell proliferation activity was significantly increased, and the apoptosis rate was significantly decreased. Conclusion The low expression of MTSS1 in bladder cancer can promote bladder cancer cell proliferation and inhibit apoptosis.
