为了确定TRH对垂体5'—脱碘酶的作用,给甲状腺功能正常及低下的大鼠腹腔注射TRH,每日两次,连续三天。 以100μg TRH/200g体重的剂量腹腔注射,观察TRH对甲状腺功能正常大鼠的脱敏感作用。注射后第4天处死,发现血清TSH、T_4和T_3均下降。以Kaplan氏或Visser氏改良法测定垂体微粒体5'—脱碘酶的活性。利用冷T_4为底物,放免测定所生成的T_3,同时测定垂体内两种脱碘酶,——高km(1.0μMT_4)和低km(10nMT_4)。注射TRH的大鼠高km5'—脱碘酶的Vmax降低为注射盐水组
To determine the effect of TRH on pituitary 5’-deiodinase, rats with normal and low thyroid function were injected intraperitoneally with TRH twice daily for three consecutive days. The mice were injected intraperitoneally with 100 μg TRH / 200 g body weight to observe the desensitization effect of TRH on normal thyroid function rats. On the 4th day after injection, the serum TSH, T_4 and T_3 were decreased. The activity of pituitary microsomal 5’-deiodinase was determined by the Kaplan’s or Visser’s modified method. Using cold T_4 as a substrate, T_3 generated by radioimmunoassay was assayed, and two deoxynivalent enzymes in the pituitary gland, -High km (1.0 μM T_4) and low km (10 nMT_4), were also determined. Vmax reduction in rat high km 5’- deiodinase injected with TRH was in the saline injection group
