目的探讨人胎盘间充质干细胞(PMSCs)在红景天苷作用下是否可以转化为肝细胞样细胞。方法分离培养人PMSCs,采用红景天苷和成纤维细胞生长因子4(FGF-4)联合应用对PMSCs进行诱导分化,倒置显微镜下每日观察诱导后细胞形态的变化;在诱导第7、14、21天,收获诱导细胞,免疫荧光染色检测CK18的表达,同时收集培养上清液,ELISA法检测白蛋白(ALB)浓度。结果在红景天苷和细胞因子的作用下,经过21 d的体外培养,PMSCs由长梭形变为多边形、类圆形,并具有肝细胞特异的表型和功能。免疫荧光可检测到诱导后细胞内CK18表达,培养上清液中有ALB的分泌。结论 PMSCs在红景天苷FGF-4作用下在体外能转化为肝细胞样细胞。
Objective To investigate whether human placental mesenchymal stem cells (PMSCs) can be transformed into hepatocyte-like cells under the action of salidroside. Methods Human PMSCs were isolated and cultured. PMSCs were differentiated by using combination of salidroside and fibroblast growth factor 4 (FGF-4). The morphological changes of PMSCs were observed daily under inverted microscope. After 21 days, the cells were harvested, and the expression of CK18 was detected by immunofluorescence staining. Meanwhile, the culture supernatants were collected and the albumin (ALB) concentration was measured by ELISA. Results Under the action of salidroside and cytokines, PMSCs changed from long fusiform to polygonal, round, and had hepatocyte-specific phenotype and function after 21 days of in vitro culture. Immunofluorescence can detect the expression of intracellular CK18 and the secretion of ALB in culture supernatant. Conclusion PMSCs can be transformed into hepatocyte-like cells in vitro under the action of salidroside FGF-4.
