目的研究Smad泛素化调节因子2(Smurf2)和核转录共抑制因子(SnoN)在T2DM大鼠肾组织中表达的动态变化及意义。方法建立T2DM模型,于成模8、12和16周处死。免疫组化和Western blot测肾组织Smurf2、SnoN、Tβ1和Smad2蛋白的表达;RT-PCR测肾皮质SnoN mRNA。结果 DM大鼠肾组织Smurf2蛋白表达增多(P<0.01),而SnoN蛋白的表达随病程进展逐渐减少(P<0.05),两者呈显著负相关(r=-0.71,P<0.01);SnoN mRNA在各组肾组织中的表达差异无统计学意义(P>0.05)。结论 T2DM肾病发病过程中,Smurf2蛋白的表达增加可能参与了SnoN蛋白的降解过程。
Objective To study the dynamic changes of Smurf2 and SnoN in renal tissues of T2DM rats and its significance. Methods T2DM models were established and sacrificed at 8, 12 and 16 weeks. The expression of Smurf2, SnoN, Tβ1 and Smad2 in renal tissue was detected by immunohistochemistry and Western blot. The renal cortex SnoN mRNA was detected by RT-PCR. Results The expression of Smurf2 protein in DM rats increased (P <0.01), while the expression of SnoN protein decreased gradually with the progression of disease (P <0.05), and there was a significant negative correlation between the expression of Snur protein and SnoN protein (r = -0.71, There was no significant difference in the expression of mRNA in each group (P> 0.05). Conclusion The increased expression of Smurf2 protein in T2DM nephropathy may be involved in the degradation of SnoN protein.
