咳敏合剂对咳嗽变异性哮喘小鼠IFN-γ及IL-13含量的影响

目的观察中药“咳敏合剂”早期干预治疗对咳嗽变异性哮喘小鼠肺泡灌洗液中白细胞介素-13(IL-13)及干扰素γ(IFN-γ)含量的影响。方法昆明小鼠随机分为5组,分别为正常对照组、模型组、咳敏合剂高剂量组、咳敏合剂低剂量组、阳性对照组,每组10只。除正常对照组外,其余小鼠用卵蛋白(OVA)联合氢氧化铝凝胶于第7、14天肌注致敏,第15至28天雾吸激发造模。第29天开始对模型组、咳敏合剂低剂量组、咳敏合剂高剂量组、阳性对照组进行药物干预,共干预5天。收集各组小鼠支气管肺泡灌洗液,采用酶联免疫吸附法(ELISA)测定IFN-γ及IL-13的含量,并观察模型小鼠肺组织病理。结果与模型组比较,咳敏合剂高、低剂量组及阳性对照组IFN-γ、IL-13含量差异有统计学意义(P<0.01)。与阳性对照组比较,咳敏合剂高、低剂量组IL-13含量差异有统计学意义(P<0.01)。病理检查显示,咳敏合剂高剂量组能有效减轻肺内炎症,与阳性对照组差异无统计学意义(P>0.05)。结论咳敏合剂可以有效减轻咳嗽变异性哮喘小鼠的气道炎症。 Objective To observe the effects of traditional Chinese medicine “Kemin Mixture” on the levels of interleukin-13 (IL-13) and interferon-γ (IFN-γ) in bronchoalveolar lavage fluid of cough-variant asthmatic mice. Methods Kunming mice were randomly divided into five groups, namely normal control group, model group, cough sensitive mixture of high dose group, cough sensitive mixture of low dose group, positive control group, 10 in each group. Except for the normal control group, the other mice were sensitized intramuscularly with OVA and aluminum hydroxide gel on the 7th and 14th day, and mice were induced by fog inhalation on the 15th to 28th days. On the 29th day, the model group, cough sensitive mixture of low-dose group, cough sensitive mixture of high-dose group, positive control group for drug intervention, a total of intervention for 5 days. The bronchoalveolar lavage fluid of each group was collected and the contents of IFN-γ and IL-13 were determined by enzyme-linked immunosorbent assay (ELISA). The lung histopathology of the model mice was also observed. Results Compared with the model group, the contents of IFN-γ and IL-13 in cough sensitive mixture high and low dose group and positive control group were significantly different (P <0.01). Compared with the positive control group, the content of IL-13 in cough sensitive mixture of high and low dose group had significant difference (P <0.01). Pathological examination showed that cough sensitive mixture of high-dose group can effectively reduce lung inflammation, and the positive control group, the difference was not statistically significant (P> 0.05). Conclusion Cough sensitive mixture can effectively reduce airway inflammation in cough variant asthmatic mice.
