目的 :观察富硒绿茶对人肝癌细胞株 Bel— 74 0 2诱导分化的影响。方法 :方法 采用给家兔灌服富硒绿茶 7d后 ,取其血清加至细胞培养基中进行实验 ,即血清药理学方法 ;方法 采用直接加富硒绿茶提取液至细胞培养基中进行实验。应用细胞生物学方法对处理后的 Bel— 74 0 2细胞进行生长曲线、克隆形成、AFP和 AL B的合成和分泌、Υ- GT活力的测定及细胞形态的观察。结果 :富硒绿茶及其血清能够抑制细胞生长及克隆形成 ,减少 AFP的合成和分泌 ,抑制 Υ-GT活性 ,促进 AL B的合成和分泌 ,并呈现一定的浓度和时间依赖关系。结论 :富硒绿茶能够诱导人肝癌细胞株 Bel—74 0 2细胞向正常方向分化 ,具有预防原发性肝癌发生的作用。
Objective: To observe the effect of selenium-enriched green tea on the differentiation of human hepatoma cell line Bel-74 0 2 . Methods :Methods Rabbits were fed with selenium-enriched green tea for 7 days. The serum was added to the cell culture medium for serum pharmacology. The method was used directly to enrich the selenium green tea extract to the cell culture medium for experiment. The cell growth curve, colony formation, synthesis and secretion of AFP and AL B, determination of Υ-GT activity and observation of cell morphology were performed on treated Bel-7402 cells using cell biology methods. Results: Selenium-enriched green tea and its serum could inhibit cell growth and colony formation, reduce the synthesis and secretion of AFP, inhibit Υ-GT activity, promote the synthesis and secretion of AL B, and show a certain concentration and time dependence. Conclusion : Selenium-enriched green tea can induce differentiation of Bel-74 02 cells in human hepatocellular carcinoma cell line to normal direction, which can prevent the occurrence of primary liver cancer.
