目的研究异常黑胆质成熟剂水提物总酚与顺铂、多西他赛联合对体外培养的人宫颈癌SiHa细胞增殖的影响,并探讨其作用机制。方法总酚分别与顺铂、多西他赛联合作用于SiHa细胞,采用MTT法检测细胞增殖抑制情况,倒置相差显微镜观察细胞形态,流式细胞术检测SiHa细胞Bcl-2、端粒酶表达水平。结果总酚-顺铂组、总酚-多西他赛组的细胞增殖抑制率分别明显高于顺铂组、多西他赛组(P<0.05),且细胞体积变小、变圆,可见较多脱落圆形死亡细胞,细胞Bcl-2、端粒酶表达水平显著降低(P<0.05)。结论异常黑胆质成熟剂总酚与顺铂、多西他赛联合可能通过降低Bcl-2和端粒酶表达水平而抑制SiHa细胞的增殖。
Objective To study the effects of the combination of total phenols of abalone saponin and cisplatin and docetaxel on the proliferation of human cervical carcinoma SiHa cells cultured in vitro and to explore its mechanism. Methods Total phenols were respectively combined with cisplatin and docetaxel in SiHa cells. MTT assay was used to detect the cell proliferation. Cell morphological changes were observed by inverted phase contrast microscope. The expression of Bcl-2 and telomerase in SiHa cells was detected by flow cytometry . Results The inhibitory rates of cell proliferation in total phenol-cisplatin group and total phenol-docetaxel group were significantly higher than those in cisplatin group and docetaxel group (P <0.05), and the cell volume became smaller and more rounded The number of apoptotic cells, the expression of Bcl-2 and telomerase in the cells were significantly decreased (P <0.05). Conclusions The combination of total phenol and cisplatin and docetaxel in abiotic Savda can inhibit the proliferation of SiHa cells by decreasing the expressions of Bcl-2 and telomerase.
