目的探讨外周血单核细胞(PBMC)中DcR3基因表达水平与子宫肌瘤的关系。方法 65名子宫肌瘤患者和62名健康对照者各取5ml静脉血,肝素抗凝,密度梯度离心法分离PBMC,TRIZOL法提取PBMC总RNA,用半定量RT-PCR法检测PBMC中DcR3基因的相对表达水平,并用t检验进行比较。结果子宫肌瘤患者组PBMC中DcR3基因的相对表达水平为(0.845±0.029),与正常对照组(0.140±0.003)相比有明显差异(P<0.01)。结论子宫肌瘤患者PBMC中DcR3基因的表达水平增高。
Objective To investigate the relationship between DcR3 gene expression and uterine fibroids in peripheral blood mononuclear cells (PBMCs). Methods Totally 65 patients with uterine leiomyoma and 62 healthy controls were divided into 5ml venous blood and heparin anticoagulated. The PBMCs were separated by density gradient centrifugation. The total RNA was extracted by TRIZOL method. The expression of DcR3 gene in PBMCs was detected by semi-quantitative RT-PCR Relative expression level, and compared with t test. Results The relative expression level of DcR3 gene in PBMC from patients with uterine fibroids was (0.845 ± 0.029), which was significantly different from that of normal control group (0.140 ± 0.003) (P <0.01). Conclusion The expression of DcR3 gene in PBMC of uterine fibroids is increased.
