目的通过两种γ计数器测量注射用重组人甲状旁腺素PTH(1-84)及其代谢产物PTH(1-34)体外药代实验的测量数据,找出进口与国产γ计数器在实际测量中的差异。方法用PE公司Wallac1470 WIZARD与中佳GC-1200γ计数器分别测定同一份PTH样品的cpm,分别建立标准曲线,对仪器的本底计数、相对测量效率、精密度等性能指标进行比较。结果 Wallac1470 WIZARD与GC-1200γ计数器测量PTH(1~84)cpm值的线性关系良好;PE-1470本底值为14±3 cpm、GC-1200本底值为57±33 cpm;PE-1470对~(125)I相对测量效率比GC-1200高出12%左右。结论 PE公司Wallac1470 WIZARD与中佳GC-1200γ计数器操作简单方便;PE-1470的准确、灵敏度略优,两者都适用于放射性体外药代样品的测定。
OBJECTIVE To measure the in vitro pharmacokinetics of recombinant human parathyroid hormone PTH (1-84) and its metabolite PTH (1-34) for injection by two γ counters and find out the difference between the imported and domestic γ counter in the actual measurement The difference. Methods The PE of Wallac1470 WIZARD and GC-1200γ counter were respectively used to measure the cpm of the same PTH sample, and the standard curve was established respectively. The background counts, relative measurement efficiency and precision of the instrument were compared. Results The linear relationship between Wallac1470 WIZARD and GC-1200γ counter was obtained. The PE-1470 background value was 14 ± 3 cpm and the GC-1200 background value was 57 ± 33 cpm. The PE-1470 pairs ~ (125) I relative measurement efficiency than the GC-1200 about 12% higher. Conclusion PE company Wallac1470 WIZARD and GC-1200γ counter are easy and convenient to operate. The accuracy and sensitivity of PE-1470 are slightly superior, and both of them are suitable for the determination of radiopharmaceutical sample.
