目的:研究姜黄素对胃癌SGC-7901细胞株NS基因表达的影响及凋亡诱导作用,探讨姜黄素抗肿瘤机制。方法:不同浓度姜黄素(0,10,20,40μmol·L~(-1))和不同时间(12,24,48 h)点处理细胞,利用CCK-8法检测细胞增殖抑制率,RT-PCR法检测姜黄素作用前后NS基因表达量的变化,TUNEL试剂盒法、流式细胞仪法检测细胞凋亡情况。结果:20μmol·L~(-1)以上浓度的姜黄素对体外培养的胃癌SGC-7901细胞株生长具有抑制作用并呈量效和时效关系;与对照组相比,姜黄素处理组NS基因表达量明显下降,细胞凋亡明显增加(凋亡指数>26.61%,P<0.05)。结论:姜黄素使胃癌SGC-7901细胞增殖减慢,凋亡增加,NS基因表达下降,可能是姜黄素抗肿瘤机制之一。
Objective: To study the effect of curcumin on the NS gene expression in gastric cancer cell line SGC-7901 and the induction of apoptosis, and to explore the anti-tumor mechanism of curcumin. Methods: The cells were treated with different concentrations of curcumin (0, 10, 20, 40μmol·L -1) and different time points (12,24,48 h). The cell proliferation inhibition rate was detected by CCK- PCR method was used to detect the changes of NS gene expression before and after curcumin treatment. The apoptosis was detected by TUNEL kit and flow cytometry. Results: Curcumin at a concentration above 20 μmol·L -1 could inhibit the growth of SGC-7901 gastric cancer cell line in vitro and in a dose-and time-dependent manner. Compared with the control group, the expression of NS gene in curcumin group The amount of apoptosis significantly increased (apoptosis index> 26.61%, P <0.05). CONCLUSION: Curcumin can slow down the proliferation, increase the apoptosis of NSCLC and decrease the expression of NS in gastric cancer SGC-7901 cells, which may be one of the mechanisms of anti-tumor effect of curcumin.
