【目的】优化阳春砂愈伤组织诱导方法,以提高愈伤组织诱导率。【方法】采用正交设计法,观察以未成熟及成熟种子为外植体,以及培养基中添加的不同植物生长物质对阳春砂愈伤组织诱导的影响。【结果】最佳培养基为MS+二氯苯氧乙酸(2,4-D)(0.5 mg/L)+6-苄基腺嘌呤(6-BA)(2 mg/L)+AgNO3(6 mg/L)+酸水解酪蛋白(1 mg/L),植物生长物质中2,4-D对阳春砂愈伤组织的形成影响最关键,未成熟的阳春砂种子是最佳外植体,愈伤组织诱导率最高达到65.41%。【结论】植物生长物质中2,4-D对阳春砂愈伤组织的形成不可或缺,成熟程度低的阳春砂种子更适宜作为愈伤组织诱导的外植体。
【Objective】 The method of callus induction of Populus euphratica was optimized to improve the callus induction rate. 【Method】 The orthogonal design method was used to observe the effects of immature and mature seeds as explants and the different plant growth substances added on the callus induction of P. euphratica. 【Result】 The best medium was MS + Dichlorophenoxyacetic acid (2,4-D) (0.5 mg / L) and 6-BA (2 mg / L) / L) + acid hydrolyzed casein (1 mg / L), 2,4-D in plant growth substances had the most important influence on the formation of callus. Injury tissue induced rate up to 65.41%. 【Conclusion】 2,4-D in plant growth material is indispensable to the formation of callus of C. mandshurica, and low maturity of C. mandshurica seeds is more suitable as callus-induced explants.
