为了研究桑叶浸出液对乳腺癌MDA-MB-231细胞增殖和对凋亡抑制蛋白Survivin的作用。本实验采用体外培养MDA-MB-231细胞,倒置显微镜观察桑叶浸出液作用后细胞形态变化;CCK-8法检测桑叶浸出液对MDA-MB-231细胞增殖的影响;流式细胞仪分析桑叶浸出液对MDA-MB-231细胞凋亡及周期的影响;Western Blot印迹法检测桑叶浸出液对凋亡抑制蛋白Survivin表达的影响。结果表明桑叶浸出液会引起MDA-MB-231生长形态的变化,抑制细胞增殖,增加细胞凋亡数量,且在桑叶浸出液作用后MDA-MB-231细胞的G0/G1期细胞明显高于正常组(p<0.05),同时桑叶浸出液降低凋亡抑制蛋白Survivin的表达,且表达量随着桑叶浸出液浓度的升高而降低。研究表明,桑叶浸出液能够抑制MDA-MB-231细胞的增殖,促进细胞凋亡及导致G0/G1期阻滞,同时能够降低凋亡抑制蛋白Survivin的表达。
In order to study the effect of mulberry leaf leachate on the proliferation of breast cancer MDA-MB-231 cells and the inhibitor of apoptosis protein Survivin. In this study, MDA-MB-231 cells were cultured in vitro, the morphological changes of the cells were observed under an inverted microscope; the effects of mulberry leaf leachate on the proliferation of MDA-MB-231 cells were detected by CCK-8 method; The effect of leachate on the apoptosis and cycle of MDA-MB-231 cells was detected by Western Blot. The effect of mulberry leaf leachate on the expression of Survivin was examined by Western Blot. The results showed that mulberry leaf leachate could induce the change of MDA-MB-231 morphology, inhibit cell proliferation and increase the number of apoptotic cells, and the cells in G0 / G1 phase of MDA-MB-231 cells were significantly higher than the normal (P <0.05). Meanwhile, mulberry leaf extract reduced the expression of Survivin, and the expression level decreased with the increase of mulberry leaf leachate concentration. Studies have shown that mulberry leaf extract can inhibit the proliferation of MDA-MB-231 cells, promote apoptosis and lead to G0 / G1 phase arrest, and can reduce the expression of apoptosis inhibitor protein Survivin.
