目的检测hepsin、maspin在正常宫颈、宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)及宫颈癌组织中的表达情况,探讨其与宫颈癌发生发展的关系。方法采用实时荧光定量逆转录-多聚酶链反应(Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction,QRT-PCR)和免疫组化SP法,检测正常宫颈组织、CIN及宫颈癌组织中Hepsin、Maspin的表达。结果 (1)hepsin在宫颈癌组织中高表达;(2)maspin在宫颈癌组织中低表达。结论 (1)hepsin的异常表达对宫颈癌的发生发展起促进和/或协同作用,可能扮演癌基因的角色;(2)maspin的异常表达对宫颈癌的发生发展起抑制作用,证实其扮演抑癌基因的角色;(3)联合检测hepsin/Hepsin、maspin/Maspin对宫颈癌的诊断、治疗、预后及放化疗后的病情判断有一定的参考价值,可能成为宫颈癌治疗中一个新的靶向分子或参考指标。
Objective To detect the expression of hepsin and maspin in cervical intraepithelial neoplasia (CIN) and cervical cancer and to investigate the relationship between the expression of hepsin and cervical cancer. Methods Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (QRT-PCR) and immunohistochemical SP method were used to detect the expression of Hepsin and Maspin in normal cervical tissue, CIN and cervical cancer . Results (1) Hepsin is highly expressed in cervical cancer tissues; (2) maspin is low expressed in cervical cancer tissues. Conclusion (1) Abnormal expression of hepsin can promote and / or synergize the occurrence and development of cervical cancer, which may play the role of oncogene. (2) Abnormal expression of maspin can inhibit the occurrence and development of cervical cancer, (3) Combined detection of hepsin / Hepsin, maspin / Maspin has certain reference value for diagnosis, treatment, prognosis of cervical cancer and judgment of disease after radiotherapy and chemotherapy, and may be a new target in the treatment of cervical cancer Molecule or reference indicator.
