目的观察雌激素(E2)对乳腺癌MCF7细胞中E-钙粘蛋白基因(CDH)的调控。方法使用Western blot、Real Time PCR分别在蛋白和RNA水平上观察雌激素如何影响E-钙粘蛋白。使用双荧光素酶报告系统检测雌激素如何影响E-钙粘蛋白启动子活性。结果雌激素下调乳腺癌MCF7细胞中E-钙粘蛋白的蛋白和RNA水平,并且能够下调E-钙粘蛋白的启动子活性。结论雌激素下调乳腺癌细胞中E-钙粘蛋白基因,为进一步探讨乳腺癌发病机制提供有力证据。
Objective To investigate the regulation of estrogen (E2) on E-cadherin gene (CDH) in breast cancer MCF7 cells. Methods Western blot and Real Time PCR were used to observe the effect of estrogen on E-cadherin at the protein and RNA levels respectively. Dual luciferase reporter system was used to detect how estrogen affects E-cadherin promoter activity. Results Estrogen downregulates E-cadherin protein and RNA levels in breast cancer MCF7 cells and is able to downregulate E-cadherin promoter activity. Conclusion Estrogen down-regulates E-cadherin gene in breast cancer cells, which may provide evidence for further study on the pathogenesis of breast cancer.
