目的:观察复方七芍降压片对自发性高血压大鼠(SHR)腹主动脉壁白细胞黏附浸润情况及动脉内皮细胞Es表达的影响,为复方七芍降压片治疗高血压病及防治靶器官损害提供理论依据。方法:21只7周龄雄性SHR随机分为模型组(SHR-S组)、复方七芍降压片组(SHR-G组)、缬沙坦(代文)组(SHR-D组)。另设7只WKY大鼠为空白对照组(WKY组);连续干预6周后取材,用HE染色法染色,观察腹主动脉壁白细胞的黏附浸润情况,免疫组化法检测腹主动脉组织Es的表达。结果:HE染色发现WKY组大鼠腹主动脉壁内皮完整,无白细胞黏附浸润,弹性纤维清晰,平滑肌细胞无空泡样变性;SHR-S组大鼠腹主动脉壁均有不同程度的白细胞黏附浸润,以黏附为主,内皮连续性较差,弹性纤维排列紊乱,平滑肌细胞显影模糊,部分大鼠平滑肌细胞有空泡样变性;SHR-G组大鼠腹主动脉壁均无白细胞黏附浸润,内皮连续,弹性纤维排列正常,平滑肌细胞显影清晰;SHR-D组大鼠腹主动脉壁有不同程度的白细胞黏附浸润,以中膜浸润为主,内皮尚连续,弹性纤维排列尚正常,平滑肌细胞显影清晰,无空泡样变性。免疫组化结果显示SHR-G组大鼠主动脉组织中Es表达阳性细胞数及表达量均少于SHR-S组(P<0.05),与WKY组及SHR-D组比较,差异无统计学意义(P>0.05);SHR-D组表达量低于SHR-S组(P<0.05)。结论:复方七芍降压片能有效减少SHR动脉管壁白细胞的黏附浸润,其降压及靶器官保护作用的机制可能是通过下调动脉内皮细胞Es的表达,进而减少动脉管壁白细胞的黏附及浸润,从而减轻动脉内皮及管壁的损伤发挥降压及保护靶器官的作用。

The results of immunohistochemistry showed that the number of es positive cells and the expression of Es in SHR-G group were less than those in SHR-S group (P <0.05). There was no statistical difference between WKY group and SHR-D group (P> 0.05). The expression of SHR-D in SHR-S group was lower than that in SHR-S group (P <0.05). CONCLUSION: Compound Qishaojiangya Tablet can effectively reduce the adhesion and infiltration of leukocytes in the arterial wall of SHR. The mechanism of antihypertensive effect and target organ protection may be through down-regulating the expression of Es in arterial endothelial cells and decreasing the adhesion of arterial wall white blood cells Infiltration, thereby reducing damage to the arterial endothelium and wall play a role in antihypertensive and target organs.
